You'd probably get more attention posting in the FITA section of AT, but whether or not that resulted in a substantive answer is another question altogether. I believe that some of the older W&W manuals said you could adjust tiller while strung, but they don't mention it now. Hoyt used to advertise that you could do both tiller and alignment when strung (when they brought the GMX out) but I suspect the liability lawyers have struck and the current manual says specifically to destring before adjustment.
Personally I would not adjust tiller while strung for a couple of reasons. One - you'll grind the finish off your limb roots. Two - I don't like the idea of moving a stainless thread on an aluminium one under tension. On the AL1 I might adjust alignment while strung, because they actually used sensibly sized grub screws with plenty of thread bearing area.
